Drugs not suitable for controlled release. Biological half-life, short half-life, and effective drugs with limited therapeutic index

There are some medicines that are not suitable for the exact release formulas. Typical features include: short biochemical age, long biological half life, potent drugs with low, high-dose, low-absorption, low-solubility, low absorption, active absorption, and activity time not exactly like rotational drug levels, .
Medicines with a controlled dosage of liberation that have a biological half-life of less than two hours or that are applied in large doses need to contain a large amount of the drug in a prohibitively prohibited manner. Medications with a long biological half life (more than 8 hours) usually provide enough long-term release in the body at normal doses. The addition of soluble compounds to controlled release formulas is not positive because melting is the always-defined step. The application of drugs such as warfarin, which has long-term pharmacological effects, does not provide any therapeutic benefits.
The combination of these compounds as some antibiotics from the beta-lactam group, fluorouracil and some amino acids that appear to absorb primarily from the intestinal microorganisms will reduce their efficacy and will provide little or no lasting effect. As previously mentioned, if the drug passes the first hepatic passageway to normal dose satisfaction, its systemic availability may be reduced after the controlled release.
Although the above discussion has been provided with useful general rules, there are many exceptions. The biologic half-life of nitroglycerin is less than half an hour. It is considered as a weak absorption drug and its liver metabolism is rapid through the first pass. However, a large number of seized products are marketed with nitroglycerine. The low circulating levels of nitroglycerin resulting from these products appear to provide adequate prophylaxis prevention of angina attacks but not acute angina seizures. Table 1-1 shows many of the dosage forms controlled by the currently marketed liberalization.
